When you look at the S5 display of the materials loaded in each material station bay, does it recognize that PVA is loaded?
I should first confirm you are using Ultimaker brand PVA which has the RFID feature on the side of the reel? Circular Tag.
If you press the button to eject, then remove the PVA reel entirely, then re-install it, and insert, the display should spin and then eventually recognize the reel.
I have not personally been successful loading a reel manually, but did not try too hard either. I have always been successfull if I just insert the filament and let the material station auto detect. If it fails, try again.
It would seem your problem is just with material recognition in the Material station, and the RFID solution can be touchy. It is not even getting to the point of loading to extruder 2. It just knows from Cura, that extruder 2 needs PVA. Probably not mechanical or related any particular bays. Maybe a bad RFID tag on reel.
