I want to extrude the filament for a certain distance for the calibration of the extruder of the nycubic i3 Mega. I tried the G1 E100 F100 and it failed

    M109 S200 succeeded, G1 E100 F100 failed

Are you sending it messages in a way that you can get responses? If you can, post any responses you get from it here.

It's also worth noting that Marlin uses speeds in millimetres per minute so the problem could be the speed. Try G1 E100 F1200 which is 20mm/s. If that doesn't work try F2400.

The USB printing feature in Cura is deprecated and no longer updated. You could try running more fully featured software designed for this sort of thing like OctoPrint which has a much more robust terminal.

Also, I'm not familiar with your printer, but almost any printer running Marlin should have the ability the control the extruder position using the printer's control panel. You may be best off using that if you can so you don't have to mess with gcode if you can avoid it.
